plasmodium berghei anka infection in icr mice as a model of cerebral malaria

چکیده
b ackground: animal models with various combination of host-parasite have long been employed to study malaria pathogenesis. here, we describe the combination of plasmodium berghei anka infec- tion in inbred icr mice as a model of cerebral malaria (cm). m ethods: infection in mice was initiated by intraperitoneal injection of 2 x 107 (0.2ml) parasitized red blood cells (prbcs). r esults: this model can produce a severe degree of infection presented by the high degree of parasitaemia followed by death 6-7 days post infection. severe anemia, splenomegaly, hepatomegaly and discolourations of major organs were observed. histopathological findings revealed several i m- portant features mimicking human cm including, microvascular sequestration of prbcs in major organs, particularly in the brain, hypertrophy and hyperplasia of the kupffer cells in the liver, pulmo- nary edema and hyaline membrane formation in the lungs and haemorrhages in the kidney’s medulla and cortex. proinflammatory cytokines tnfα, ifnγ, il-1, il-6 and il-18, and anti-inflammatory cytokine il-10 were all found to be elevated in the plasma of infected mice. c onclusion: this model can reproduce many of the important features of cm and therefore can be used as a tool to advance our understanding of the disease pathogenesis.

Plasmodium berghei ANKA Infection in ICR Mice as a Model of Cerebral Malaria
BACKGROUND Animal models with various combination of host-parasite have long been employed to study malaria pathogenesis. Here, we describe the combination of Plasmodium berghei ANKA infection in inbred ICR mice as a model of cerebral malaria (CM). METHODS Infection in mice was initiated by intraperitoneal injection of 2 x 10(7) (0.2ml) parasitized red blood cells (PRBCs). RESULTS This mode...

In this paper, we have collected the findings of available literature focusing on brain metabolites by spectroscopy in the murine model of cerebral malaria disease. The literature search for experimental cerebral malaria (ECM) and spectroscopy using National Institute of Health's PubMed database provided us with 9 peer-reviewed publications. These publications have used mice infected with Plasm...
متن کاملPrevention of experimental cerebral malaria by Flt3 ligand during infection with Plasmodium berghei ANKA.
Dendritic cells are the most potent antigen-presenting cells, but their roles in blood-stage malaria infection are not fully understood. We examined the effects of Flt3 ligand, a cytokine that induces dendritic cell production, in vivo on the course of infection with Plasmodium berghei ANKA. Mice treated with Flt3 ligand showed preferential expansion of CD8(+) dendritic cells and granulocytes, ...
متن کاملCharacterization of cerebral malaria in the outbred Swiss Webster mouse infected by Plasmodium berghei ANKA.
Plasmodium berghei ANKA (PbA) infection in susceptible inbred mouse strains is the most commonly used experimental model to study pathogenesis of cerebral malaria (CM). Indeed, many concepts on mechanisms related to this complication have arisen from works using this model. Although inbred strains present several advantages and are indicated for most studies, the use of outbred models can show ...

Pathogenesis of Plasmodium berghei ANKA infection in the gerbil (Meriones unguiculatus) as an experimental model for severe malaria
BACKGROUND As the quest to eradicate malaria continues, there remains a need to gain further understanding of the disease, particularly with regard to pathogenesis. This is facilitated, apart from in vitro and clinical studies, mainly via in vivo mouse model studies. However, there are few studies that have used gerbils (Meriones unguiculatus) as animal models. Thus, this study is aimed at char...
